Letrozole is a potent and highly selective, reversible, third-generation, nonsteroidal aromatase inhibitor. Originally developed as an antineoplastic drug for the treatment of breast cancer, aromatase inhibitors are commonly used to lower estrogen levels and increase testosterone production in men. By inhibiting the aromatase enzyme, letrozole prevents the conversion of testosterone to estrogen, thereby reducing the negative effects of excess estrogen on the brain, hypothalamus, and hormone secretion.

Letrozole cannot reduce already elevated estradiol levels, but it can prevent further estrogen formation.

Purpose of Use

In bodybuilding, aromatase inhibitors are primarily used during anabolic steroid cycles. It’s important to note that not all anabolic steroids undergo aromatization. Letrozole is typically used in combination with testosterone and its derivatives, such as methandrostenolone and methyltestosterone.

Moderate estrogen levels are beneficial for muscle growth, mood, and libido, so prophylactic dosages are recommended to maintain the necessary estrogen balance in men.

How to Use

An optimal approach is to monitor estradiol levels every 10–14 days when using anabolic steroids with short half-lives (e.g., testosterone propionate, methandrostenolone) or after 3–4 weeks with long-acting esters (e.g., enanthate, Sustanon, cypionate). Based on the results, a starting dose of letrozole of 0.25 mg daily or every other day is usually prescribed. After 10 days, another estradiol test is performed and the dosage is adjusted accordingly.

An effective letrozole dosage starts at just 0.02 mg, which is 100 times lower than the therapeutic dose. This dose can reduce estrogen levels by about a third. Letrozole has a half-life of 2–4 days, allowing for administration every other day. It is crucial to choose the dosage based on blood test results to avoid excessive suppression of estrogen levels.
